Type 2 immune responses – aspects in infection, tissue remodeling, cancer and allergy

Contents The first part of the course will cover all cellular mechanisms, which are involved in the induction of type 2 immune responses. This includes sensing and uptake of agents such as parasite molecules, allergens or other foreign bodies by epithelial cells, macrophages or dendritic cells as well as the activation of these cells. Further, the molecular processes of T-cell differentiation and activation (both TH2 cells and Tregs) will be covered. The roles of ILCs, Mast cells, basophils, eosinophils and M2 macrophages in type 2 inflammation in various tissues and disease settings (infection, allergy cancer, fibrosis) will be discussed in detail and a variety of effector mechanisms (surface molecules, cytokines, chemokines, lipid mediators) will be explained in these contexts. Mouse models commonly used to study type 2 immune responses (e.g. allergy and helminth infection) will be discussed. Aspects of age, microbiome influence and maternal imprinting will also be covered in several of the lectures. Furthermore, clinical aspects such as pharmaco- and immunotherapy as well as State of the Art molecular techniques will be discussed.
Didactic Aim/Competences - Deep mechanistic understanding of type 2 immune responses (from epithelial sensing and alarmin production via dendritic cell activation, TH2/ ILC2 activation, to the activation of effector cells (granulocytes, Mast cells,...) and epithelial/ fibroblast remodeling) - Understanding of the clinical relevance of type 2 immune responses in various disease settings such as allergies, helminth infection, fibrosis and cancer as well as the functional and mechanistic differences between these diverse disease settings - Acquire knowledge about biomarkers and therapeutic strategies in type 2 inflammation in different diseases - Understanding of tissue specific aspects of type 2 immune responses (skin, upper and lower airways, liver, intestine)
